Pustulate Ascomycete on Sambucus
Peter Thompson, 19-01-2013 11:54
Hello.

I have an ascomycete found growing on decorticated wood of Sambucus. Blackish pustules are to be seen at the surface, each covering about five embedded perithecia arranged in a circle.
The wood surrounding the pustules is clearly discoloured yellow. Although the spores are a golden yellow colour, I can see no evidence of extruded spores being responsible for the yellowness under the microscope.
I have attached a sketch of the microscopy, including a horizontal section through the perithecia. The spores seem to be pseudoseptate with similarities to those of the genera Prosthecium and Pseudovalsa.
Also attached is a photograph showing the pustules and yellow discolouration around them.

I wonder if anyone has any idea about the identification of this fungus.

Thank you,
Peter.

Hello Peter, at first sight I would suggest the genus Lepteutypa but I am not aware of a species staining the wood yellow. You should take a look at Barr 1994, Mycotaxon 51, pp 191-224.
Good luck,
Jacqques

Re : Pustulate Ascomycete on Sambucus
Hello Jacques,

Thank you for your reply.

I am trying to locate the Barr 1994 document on the Internet, but have so far only been able to see it listed as a reference in documents written by others.

Do you know where I should be able to find the text of the document itself?

I also notice from illustrations of microscopy on the Internet, that other species in the genus Lepteutypa have appendages on their spores. Do you know whether there are species in the genus without appendages?

Thank you,
Peter.

Re : Pustulate Ascomycete on Sambucus
Hello Peter,
you can access all older paper published by Mycotaxon through Cyberliber.
Ascospores in Lepteutypa do not have appendages but some of them are said to have a gelatinous sheath. They are distoseptate and usually show dark granulations encrusting the septa, something to check on your material
I found no good match of your collection with species listed by Barr. Lepteutypa hederae has similar ascospores but its apical ring is not amyloid and the substrate is not discoloured.
I will send you separately the paper by Shoemaker and Müller dealing with L. hederae for your information.
The funny thing I also have a nice collection of Lepteutypa on Sambucus with five septate ascospores, different from yours and likewise unnamed!
Cheers,
Jacques

Re : Pustulate Ascomycete on Sambucus
Hello Jacques,

I do have access to Cyberliber, which I shall bookmark for future reference.

I have found the paper which you named and will have a look through it, as well as some of the other information which is available there.

Thank you,
Best wishes,
Peter.

Dear Peter,
could you be so kind to send me a good part of the material for culture preparation.
Thanks,
Walter
