Induction of ascomata in pure culture
Björn Nordén, 07-01-2019 12:40
Hi,

I wonder if anyone has experience of induction of ascomata in pure culture? I suspect the methods are fairly species-specific or are there some protocols involving various media, temperatures and light et c that can be used for screening a lot of various ascomycetes? I found these articles so far:

Houbraken J, Dyer PS (2015): Induction of the Sexual Cycle in Filamentous Ascomycetes... In: van den Berg, MA., Maruthachalam, K (Eds.) (2015): Genetic Transformation Systems in Fungi, Volume 2 (Fungal Biology). Springer. DOI: 10.1007/978-3-319-10503-1_2.

Untereiner Wendy A. (1994): A Simple Method for the in Vitro Production of Pseudothecia in Species of Capronia. Mycologia 86, No. 2 (Mar. - Apr., 1994), pp. 290-295

Grateful for any advice! (for example, lab, person, articles).

Hello Björn,

Maby the simplest way to find these things out is to write an e- mail to the Westerdijk-Institute.

I had good advices from the kitchen there, that time, they are an open institute and are willingly to help as much they can, is my expierence.

Otherwise there are some, antiquarian books, but the same Westerdijk-institute has a manual about tese things.

Best,

Marco

Excellent idea! Thank you!

Re : Induction of ascomata in pure culture
Two rather brief general hints here:

1. adding the appropriate sterilized substratum to the agar plate (bark, stem, whatever you isolated from) helps to provide a physical base on which ascomata can develop.

2. Don't overfeed your fungus - culture media with high nutrient levels encourage vegetative growth only. Fungi often produce fruit bodies when stressed, when they "think" that their food source is running out.

But there are about as many methods as there are species, so experimentation is inevitable. And do be patient, sometimes fruit body formation can take months...

Best wishes

Paul
