Which asco is this?
Joop van der Lee, 06-08-2012 21:00

Hello all,

I am anxious about the name of these ascomycetes. Growing on a fallen trunck of a maple. The colours speak for themselves. Spores long and sometime bended 6.66-8.05x1.63-1.91 um an can contain up to 3 droplets, parafyses 31.59x1.59 um with rounded top, asci containing 8 spores 36.77-36.84x4.45-4.81 um. Sorry about the poor quality of the micro foto's but they were processed by  a laptop and not by my desktop.
The first two foto's of the asco were taken 30062012 and the next two at 12072012.

Joop

Re : Which asco is this?
Ionomidotis fulvotingens. Asci should be inamyloid, and adding KOH to a water mount must extrude a yellow-brown sap.

Re : Which asco is this?
Thanks for the quick reaction Zotto. Is the asci maintaining his predominant green colour or is it turning black in an older stage? The colour change is what I have seen on the internet for this typeof asci.

I will try what the reaction will be with KOH.

Re : Which asco is this?
The fungus actually turns black with age, yes, and especially when dry (the fungus is drought-tolerant). After rehydrating the colour gets more greenish again, but not as light as when younger.

Re : Which asco is this?
Hans-Otto I tried pure KOH 5% on the asco and it turned black also when mixing KOH with water it turned black. No yellow-brown sap extraction observed. Did I do something wrong?

Re : Which asco is this?
Did you test it by macroscopy? or in a water mount under the microscope? The latter alloews to see the pigment extruding in the medium. I never tried to check the colour change by putting directly on the apothecia. Can you show us a photo of your result?

Hello Hans,

Here you have a photo of the colour change in black using 5% KOH on the apothecia, working on the microscopic photo.

Re : Which asco is this?
Thanks, Joop, this is almost incredible. the next time I will check KOH also on macro. Did you also test a fragment in water under the microscope with KOH? It should release a dye.

Re : Which asco is this?
The person who initially found the asco will do that tomorrow Hans for it is in her area and quit a distance from where I live in Holland.

Re : Which asco is this?
On the left you see an untypical Ionomidotis ?fulvotingens with a honey-yellow dye extruding in KOH. Right a normal I. fulvotingens with a more red-brown dye. I suspect your fungus shows the latter colour. The left one is by Alex Akulov and was a matter of discussion already here, though I cannot trace it.

Re : Which asco is this?
Helloo Hans,

Here is the result of the microscopic research.
Foto-1 is taken with water only and foto-2 is taken with water and 5% KOH.
Does this confirm that it is I. Fulvotingens?

Re : Which asco is this?
Yes, it does. The photos are a bit overexposed. It seems that the yellow dye dissolves in the medium already ?in water, and that KOH provokes a red reaction.

I feel I need to study this reaction more detailed in the future, particularly because there might be two species involved, and because it is name-giving for the ionomidotic reaction.
